Considering you mentioned "quiver" I'm assuming you're a ranger. Ranger isn't my main class, so I won't comment very much about gear choices or skill trees.

As for what to transfer onto each item: That would depend on the base stats/unique value on each item. Use your brain for this one, don't just blindly follow whatever.

Gems: Anywhere. Higher difficulty is more rewarding, as are bosses. As for which ones to turn into dust - I think most people are converting armor and resistance gems into dust.

Runes: I believe difficulty probably does affect drop rate, but I doubt we'll ever pry that info out of BP. That said, difficulty mode does affect the rarity of the rune you can get. I think merciless can drop up to green rarity, and bloodshed up to blue rarity.

About wisdom, I made a guide about it a few months ago. The only addition I have to make about it is that the Chamber of Panoltiaco (accessed from Nahuatlan) is a great place to farm wisdom since you get to smack a Sentinel and a Boss one right after the other. You do have to be able to survive Infernal mode tho. :p
